葛藤を生じる意思決定における前頭前野・扁桃体基底外側核の機能

Prefrontal cortex(PFC)and basolateral nucleus of amygdala(BLA)are involved in systems of reward and punishment, which are potent motivation for action. Here, we investigated the role of PFC and BLA in decision making under conflict by using conflict task, in which outcomes of the action are both reward and punishment. In conflict task, rats received one pellet and electric footshock simultaneously after lever-pressing during tone presentation. The current intensity of footshock became stronger each time the rats pressed lever 10 times, and rats were allowed to choose whether to press lever or not. Reversible inactivation of the ventral-medial PFC(v-mPFC)or BLA by local treatment with GABAA and GABAB agonists elevated maximum current intensity(MCI), in which lever press probability is more than 25%, whereas MCI was lowered by ventral-lateral PFC(v-lPFC)inactivation. Although, the probability of lever press for only reward(no-conflict state)is not affected by inactivation of all of three regions, latency to lever press was increased by v-lPFC and BLA inactivation. Unit recording during conflict task and tones which predict reward(pellet)or punishment(footshock)revealed that large population of v-lPFC neurons were excited by tone preceding lever press in conflict task and reward predictive tone. Many BLA and v-mPFC neurons were excited by tone preceding lever press, tone during conflict, reward or aversive predictive tones. These observations suggest that v-lPFC contributes to reward-seeking, whereas v-mPFC and BLA contribute to both reward-seeking and punishment-avoidance in decision making under conflict. Further, v-mPFC and BLA have function in conflicting state, indicating that these regions might be involved in thinking process under conflict.

報酬予測と行動調節を担う扁桃体-海馬間のオシレーション相互作用

予測される報酬によって、我々の行動は促進されたり、逆に抑制されたりする。この過程は、それぞれ固有の情報処理を担う異なる脳部位が互いに情報をやり取りすることによって実現される。しかし、このような報酬予測による行動調節を担う部位間の相互作用に関しては、未だ不明な点が多い。この相互作用を検出するうえで最も有力な指標は、異なる部位間で生じるオシレーションの同期であるが、報酬予測と行動調節それぞれ関わる部位間の相互作用をオシレーションの同期として検出し、それがどのような機能を果たしているのか直接検討した研究はまだない。そこで本研究は、報酬が提示される確率の高低を予測しながら弁別課題を遂行しているラットの扁桃体と海馬からニューロン活動と局所場電位(LFP)を同時に記録し解析した。その結果、以下の5点が示された。第一に、報酬期待が高いとき、ラットは有意に速い反応時間と高い正答率を示した。第二に、扁桃体からは報酬の期待に、海馬からは課題中の行動調節に関連するニューロンがそれぞれ有意に多く記録された。第三に、報酬期待が高いとき、扁桃体と海馬の間において、LFPが示す高周波数オシレーション(HFO;90-150 Hz)の同期が増加した。第四に、報酬期待が高いときにのみ、この同期は課題中の反応時間と有意な相関を示した。第五に、報酬期待が高いとき、扁桃体HFOのパワーはthetaリズムによって特異的に変調していた。これらの結果は、扁桃体において報酬の予測が、海馬において課題に必要な行動の制御が、それぞれ担われており、それらの情報が高周波オシレーションの同期によりやり取りされていることを示唆している。今後は、前頭皮質や線条体などの部位も含めたより広範な回路において、オシレーションの同期の機能を検討していく必要があるだろう。

痛みによる負情動生成の神経機構

痛みは、侵害刺激が加わった場所とその強さの認知に関わる感覚的成分と侵害受容に伴う不安、嫌悪、抑うつ、恐怖などの負情動の生起に関わる情動的成分からなる。痛みによる負情動は、生体警告系としての痛みの生理的役割にとって重要であるが、慢性疼痛では、痛みにより引き起こされる不安、抑うつ、恐怖などの負情動が、患者のQOLを著しく低下させるだけでなく、精神疾患・情動障害の引き金ともなり、また、そのような精神状態が痛みをさらに悪化させるという悪循環をも生じさせる。しかしながら、慢性疼痛による情動機構の可塑的変化のメカニズムはおろか、痛みによる負情動生成の神経機構についてもほとんどわかっていないのが現状である。我々は、痛みによる負情動生成機構について、「extended amygdala」を構成する脳領域である分界条床核に着目して研究を進めている。本講演では、背外側分界条床核内CRFおよびNPY神経情報伝達が痛みによる負情動生成を相反的に制御していることを示す行動薬理学的および電気生理学的研究の成果を示すとともに、分界条床核から腹側被蓋野に投射する神経経路に関する組織学的解析の結果を紹介し、痛みによる負情動生成の神経機構について論じたい。

マウント及び射精に対するオスラット側坐核の報酬関連応答

 報酬に対する側坐核のニューロン応答は主に食物報酬を用いて研究されており、性的報酬に関して側坐核のニューロンがどのような応答を示すのかは不明であった。そこで本研究では、性行動中のオスラット側坐核からニューロン活動を記録した。記録したニューロンは、電気生理学的特徴(スパイク波形、平均発火頻度、および不応期の長さ)に基づき、中型有棘ニューロン、高発火性抑制性ニューロン(パルブアルブミン陽性介在ニューロン)、大型無棘ニューロン、およびその他のニューロンの4つのいずれかに分類した。次にこれらのニューロンのマウントの前後における発火頻度の変化、および射精の前後での周期的活動(オシレーション)を解析した。
 合計123個のニューロンを記録し、この内、60個が中型有棘ニューロン、21個が高発火性抑制性ニューロン、6個が大型無棘ニューロン、36個がその他のニューロンに分類された。マウントに対する応答性の解析では、イントロミッション(ペニスの挿入を伴うマウント)に対して多くの高発火性抑制性ニューロンが抑制性の応答を示すこと(応答したニューロンのうち75%)が明らかになった。先行研究でも、高発火性抑制性ニューロンが食物報酬に対して抑制性応答を示すことが報告されている。射精前後でのオシレーションの解析では、射精後に射精前に対して約3倍、デルタ帯域および高ガンマ帯域でオシレーションを呈するニューロンの数が増加することが明らかになった。食物報酬を用いた先行研究により側坐核における高ガンマ帯域のオシレーションは報酬知覚に関与していることが報告されている。また、快感のメカニズムに関与するオピオイド受容体の作動薬は、ヒトでオルガズムに似た作用を持ち、デルタ帯域のオシレーションを増加させることが報告されている。
 以上から、いくつかのメカニズムが性的報酬と食物報酬とに共通していることが示唆された。本研究結果は、報酬系の普遍的・本質的な機能の理解や報酬系の機能異常がもたらす性機能障害の理解に役立つと考えられる。

Discovery of glutamatergic neurons intermix with dopaminergic neurons in the substantia nigra and ventral tegmental area

Midbrain dopamine systems play important roles in Parkinson's disease, schizophrenia, addiction, and depression. The participation of midbrain dopamine systems in diverse clinical contexts suggests that these systems are highly complex. It is well established that in addition to dopamine neurons, the midbrain dopamine areas contain GABAergic neurons. Here, we present evidence indicating that the midbrain dopamine area has also glutamatergic neurons. Vesicular glutamate transporter 2(VGluT2)mRNA-expressing neurons are observed within each midbrain dopamine area. Within the retrorubral field(RRF), large populations of VGluT2 neurons are observed throughout its anteroposterior extent. Within the substantia nigra pars compacta(SNC), VGluT2 neurons are observed centrally and caudally, and are most dense within the laterodorsal subdivision. RRF and SNC rat VGluT2 neurons lack tyrosine hydroxylase(TH), making them an entirely distinct population of neurons from dopaminergic neurons. The ventral tegmental area(VTA)contains the most heterogeneous populations of VGluT2 neurons. VGluT2 neurons are found in each VTA subnucleus but are most dense within the anterior midline subnuclei. Some subpopulations of VGluT2 neurons co-express either TH or glutamic acid decarboxylase(GAD). Different subsets of rat VGluT2-TH neurons exist based on the presence or absence of vesicular monoamine transporter 2, dopamine transporter, or D2 dopamine receptor. Thus, the capacity by which VGluT2-TH neurons may release dopamine will differ based on their capacity to accumulate vesicular dopamine, uptake extracellular dopamine, or be autoregulated by dopamine. VTA VGluT2 neurons exhibit intrinsic VTA projections and extrinsic projections to the accumbens and to the prefrontal cortex. Given their molecular diversity and participation in circuits involved in addiction, we hypothesize that individual VGluT2 subpopulations of neurons play unique roles in addiction and other disorders.
