Lysophosphatidic acid(LPA)はラットC6グリオーマのネクローシスを誘導し生存率を低下させる

これまでに我々は、選択的セロトニン再取り込み阻害薬(SSRI)であるセルトラリンを3週間慢性投与したラットの脳内で発現変化する遺伝子を探索し、Lysophosphatidic acid(LPA)シグナル伝達系がうつ病の回復過程に関与するという仮説を提唱している。(Yamada et.al. 2002)。またLPAと情動の関連について明らかにするため、マウス脳室内にLPAを投与し情動行動変化を検討したところ、LPAはLPA受容体(LPAR;7回膜貫通型G蛋白共役受容体)を介して不安を惹起することが明らかとなり、情動の制御において何らかの役割を担っているものと推察された。一方、情動調節にけるグリア細胞の果たす役割が近年注目を集めている。そこで、今回我々は、培養細胞モデルとしてラットC6グリオーマ(C6細胞)を用いて、LPAがグリア細胞に与える効果を検討した。まずC6細胞がLPAR1-6を発現していることをRT-PCR法により明らかとした。次に、MTTアッセイによる細胞毒性試験を行い、LPAがC6細胞の生存率や増殖率に与える影響を評価した。その結果、通常の培地で飼育したC6細胞ではLPAは生存率や増殖率を変化させなかったが、血清不含培地で飼育したC6細胞では培地中のLPA濃度依存的に生存率が低下することが確認された。しかし、LPAR阻害作用をもつBrP-LPAの事前添加によっても、この生存率低下効果が抑制されず、この細胞毒性はLPARを介した作用ではない可能性が示唆された。続いて、この生存率低下効果がLPAの分解により生成された不飽和脂肪酸が培養細胞に与えた酸化ストレスに起因するものと仮定して強力な抗酸化作用を有するアスコルビン酸を事前添加し実験を行ったが、アスコルビン酸は細胞活性の低下に影響を示さなかった。そのため、LPAの細胞毒性は酸化ストレスを介したものではない可能性が示唆された。一方、エチジウムホモダイマーIII、FITC-アネキシンV、Hoechst 33342を用いてC6細胞を染色し顕微鏡観察を行ったところ、血清不含培地で培養したC6細胞にLPAを作用させた群ではFITC-アネキシンVで染色される細胞の割合が増加する傾向が見られた。このことからLPAはLPARを介する経路やそれ自身が与える酸化ストレス以外の機構を介して、アポトーシスではなく、ネクローシスを誘導しており、その結果、細胞生存率が低下する可能性が示された。LPAは両親媒性であり、界面活性作用を持つ。今回発見されたLPAの予想外の作用は、この両親媒性に由来するものであるかも知れない。

ドネペジルのミクログリアに対する作用について

アルツハイマー型認知症(AD)治療においてアセチルコリンエステラーゼ(AchE)阻害剤であるドネペジルは認知機能障害の進行を遅らせる。神経細胞に対するドネペジルの作用機序として、AchE活性抑制効果のほか、脳虚血やグルタミン酸による神経毒性に対する神経保護作用があり、nAChα7受容体あるいはsigma-1受容体を介する機序や、細胞内Ca濃度([Ca2+]i)上昇を抑制する効果、PI3K経路活性化などが関与するとされている。またADの病態にミクログリア活性化が関与するとの報告が相次いでいるが、ドネペジルのミクログリアに対する効果については、Aβオリゴマー負荷によるTNFα,IL-1β,およびNO等の産生を抑制するとの報告や、LPSあるいはATP刺激後の[Ca2+]iの上昇を、sigma-1受容体を介して抑制するとの報告があるのみであり、関与する細胞内機序についての報告はない。今回我々は、ドネペジルのミクログリアへの作用機序について,細胞内カルシウムシグナリングに主に着目して検討した。ドネペジルを前処置したミクログリアおいては、TNF投与後の[Ca2+]i上昇が抑制されたが、細胞内機序として、sigma-1受容体、PLC系およびMAPK系は関与しなかったが、PI3K/Akt経路が関与した。AD治療において、ドネペジルは主にPI3K/Akt経路を介してミクログリア活性化を制御する可能性が示唆された。

Reactive astrocytes observed in the brain of dystonia musculorum mice

Dystonia musculorumdt)マウスは、細胞骨格制御因子をコードすると考えられているdystonin遺伝子の変異により感覚神経変性および、ジストニアや小脳失調などの運動障害が生じるミュータントマウスである。dtマウスの感覚神経や脊髄の一部のニューロンでは、ニューロフィラメント(NF)の異常な蓄積が報告されている。また、さまざまな神経変性疾患で、NFの異常な蓄積が報告されている。今回、我々が作成したdystonin遺伝子トラップマウス(DstGtホモマウス)の脳全体におけるNFの蓄積を調べるとともに、グリア細胞の一つであるアストロサイトについても、組織学的に検討した。その結果、NFの蓄積は末梢の感覚神経系のみならず、中枢神経系においても広く観察された。特に軸索に蓄積するスフェロイドは、脳全体において観察された。NFの細胞体における蓄積像は、主として脳幹の運動や感覚の情報伝達に関わる神経核(前庭神経核および網様核大細胞部など)において観察された。さらに、これらの神経核周囲において、GFAP強陽性の反応性アストロサイトが観察された。アストロサイトの機能分子の発現について調べたところ、水チャネルのAquaporin mRNAが発現低下していることがわかった。以上の結果から、dtマウスではNF蓄積ニューロンの機能異常により運動障害が生じる可能性と、脳内の水バランス制御に異常があることが示唆された。

Simvastatin regulates expression of neprilysin in cultured astrocytes

 One of the major neuropathological hallmarks of Alzheimer's disease is the deposition of amyloid β-protein(Aβ)in the brain. The accumulation of Aβ seems to arise from an imbalance between Aβ production and clearance. Numerous studies indicate that deficits in the brain expression of neprilysin(NEP)and insulin-degrading enzyme(IDE), the important Aβ-degrading enzymes in the brain, may promote Aβ deposition in patients with sporadic late-onset AD. Statin, which are used clinically for reducing cholesterol levels, can exert beneficial effects on AD. Therefore, we examined whether various statins are associated with Aβ degradation by inducing NEP and IDE expression, and then have relation on the signal transduction of between its induction, inhibition of cholesterol production, and morphological change. The treatment of simvastatin and atorvastatin on the cultured rat astrocytes significantly decreased the expression of NEP, but not IDE, with the concentration- and time-dependent through the activation of extracellular signal-regulated kinase(ERK). This reduction of NEP expression was achieved by the release into the extracellular of cultured astrocytes. Furthermore, the cultured medium prepared from simvastatin- and atorvastatin-treated astrocytes significantly induced the degradation of exogenous Aβ. These results suggest that simvastatin and atorvastatin induce the increase of Aβ degradation of NEP on the extracellular of astrocytes by inducing ERK-mediated pathway activity, and that these reagents regulate the differential mechanisms on between the secretion of NEP and the induction of cholesterol reduction and morphological changes in the cultured astrocytes.

Functional analysis of DISC1 binding protein DBZ in oligodendrocyte differentiation

Schizophrenia(SZ)is a serious and disabling mental disorder with a lifetime prevalence of about 1% of the population worldwide, and commonly has a chronic course. The underlying pathological mechanisms are still largely unknown, but a growing body of evidence suggests that it is a multifactorial disorder influenced by genetic, neurodevelopmental and social factors. Disrupted-in-schizophrenia 1(DISC1)is a gene disrupted by a(1;11)(q42.1;q14.3)translocation that segregates with major psychiatric disorders including schizophrenia, recurrent major depression and bipolar affective disorder in a Scottish family. Previously, we reported that DBZ(DISC1 Binding Zinc-finger protein), a brain-specific member of DISC1 interactome, is expressed in oligodendrocyte lineage cells, and positively regulates oligodendrocyte differentiation in vivo and in vitro. To elucidate the role of DBZ in oligodendrocyte differentiation, we focus on known DBZ interactors, Nde1(nuclear distribution gene E homolog 1)and NdeL1(nuclear distribution gene E-like homolog 1). To examine if Nde1 or NdeL1 is expressed in oligodendrocyte lineage cells, we performed in situ hybridization combined with immunohistochemistry analysis of mouse corpus callosum. At P14, the period of myelination, DBZ mRNA is highly expressed in CC1 positive oligodendrocyte lineage cells in mouse corpus callosum. Nde1 mRNA is expressed in CC1 positive oligodendrocyte lineage cells, but not in PDFGR positive oligodednrocyte precursor cells. As multiple lines of evidence obtained by brain imaging, studies in postmortem brains and genetic association studies have implicated oligodendrocytes and myelin dysfunction in SZ, these results may provide important clue about the underlying etiology of SZ.

Extracellular vesicles from vascular endothelial cells affect the behavior of oligodendrocyte precursor cells

We previously demonstrated that brain microvascular endothelial cell(MVEC)transplantation promoted remyelination of demyelinated axons in the infarct induced in the internal capsule of adult rat brain by stereotactic injection of endothelin-1 and reduced apoptotic death of oligodendrocyte precursor cells(OPCs)in and around the infarct. We also found that the conditioned medium(CM)from cultured MVECs inhibited apoptosis of cultured OPCs. In this study, we examined contribution of extracellular vesicles(EVs)contained in the CM to its inhibitory effect on OPC apoptosis. We isolated OPCs from postnatal day 0-2 rat cerebral cortices by the immunopanning method, and cultured them in serum-free medium containing PDGF as a mitogen. MVECs were prepared from adult rat cerebral cortices, and cultured in endothelial cell growth medium. After 2 days in culture with EVs derived from MVECs, there was significantly less pyknotic and more BrdU-positive OPCs when compared to control. OPCs migrated longer in the presence of EVs when compared to control. To examine whether these effects on cultured OPCs are shared by EVs from endothelial cells(ECs), we prepared EVs from conditioned media of several types of ECs, and tested their effects on cultured OPCs. EVs from all types of ECs we examined reduced apoptosis of OPCs and promoted their motility. As identification of the molecules contained in EVs from ECs may prove helpful for establishment of effective therapies for demyelinating diseases, mass spectrometric analysis was performed to identify proteins contained in EC-derived EVs. We found several candidate proteins in EVs released from ECs which affect OPC behaviors. We are now analyzing miRNA contained in the EVs from ECs.
