トリブチルスズによる核呼吸因子-1(NRF-1)阻害を介したGluR2発現減少

【目的】トリブチルスズ(TBT)は船底塗料等に使用されてきた環境化学物質である。我々はTBTがAMPA型グルタミン酸受容体GluR2サブユニットの発現減少に伴い、神経毒性を引き起こすことを報告しているが、その詳細なメカニズムは未だ明らかとなっていない。核呼吸因子-1(NRF-1)はミトコンドリア関連遺伝子や、GluR2の発現を制御している転写因子であり、転写共役因子peroxisome proliferator-activated receptor gamma coactivator-1α(PGC-1α)との相互作用により転写活性が上昇する。本研究では、TBTがNRF-1転写活性に及ぼす影響について検討した。
【方法】実験には胎生18日齢ラット(Slc:Wistar/ST)より調製した大脳皮質初代培養神経細胞を用いた。細胞は培養6日目にAra-Cを添加しグリア細胞を死滅させ、培養11日目で各実験に使用した。20 nM TBTを3-24時間曝露し、クロマチン免疫沈降法(ChIP assay)及びゲルシフトアッセイによりNRF-1のDNA結合活性を評価した。NRF-1核移行は神経細胞より核タンパク質を抽出し、特異的抗体を用いたウエスタンブロッティングにより評価した。NRF-1-PGC-1α複合体は磁気ビーズを用いた免疫沈降法により精製し、ウエスタンブロッティングによりその形成量を評価した。
【結果・考察】ラット大脳皮質初代培養神経細胞に20 nM TBTを3-24時間曝露することによりNRF-1のGluR2プロモーターへの結合活性が低下することがChIP assayより明らかとなった。さらに、TBTを添加した細胞の核画分とNRF-1結合DNA配列を用いたゲルシフトアッセイの結果でも同様にNRF-1のDNAに対する結合活性の低下が認められた。また、TBT曝露によりNRF-1核移行に影響は与えなかったものの、NRF-1-PGC-1α複合体レベルの低下が認められた。NRF-1は神経細胞の発達や樹状突起の伸長に関与することは報告されているが、AMPA受容体サブユニットに対する影響は明らかとなっていない。本研究により、TBTはNRF-1とPGC-1αの相互作用を阻害し、NRF-1の転写活性を低下させることでGluR2発現を減少させる可能性が示唆された。

プロスタグランジンE2によるミクログリアのアポトーシスの誘導

Prostaglandin E2(PGE2)plays a critical role in the modulation of microglial function including migration and phagocytosis through EP2, which increases intracellular cyclic adenosine monophosphate(AMP)concentration. In the present study, we have found that PGE2 reduces cell viability in microglia.
PGE2 decreased MTT reduction, and increased LDH release, DNA fragmentation, and PARP cleavage after 24 h incubation, suggesting that PGE2 induces apoptosis in these cells. An EP2 agonist butaprost and an EP4 agonist PGE1 alcohol also induced apoptosis, while an EP1 agonist 17-phenyl trinor PGE2 or an EP3 agonist sulprostone at 10-6 M did not. On the other hand, EP1-EP4 antagonists, SC-51322, AH6809, L-798106, or GW627368X up to 10-5 M did not affect decrease in MTT reduction by PGE2. Intracellular cyclic AMP accumulation was induced by butaprost, but not 17-phenyl trinor PGE2, sulprostone, or PGE1 alcohol at 10-6 M. Additionally, we previously reported that PGE2-induced intracellular cyclic AMP accumulation was reversed by AH6809. These results suggest that PGE2 induces apoptosis in microglia independent of intracellular cyclic AMP concentration, and there are different mechanisms between PGE2-induced apoptosis and modulation of microglial function.

培養大脳皮質ニューロンに対するカベルゴリン(ドーパミンD2受容体アゴニスト)の神経保護効果の解析

Development of the efficient medicine for brain diseases along with neuronal cell death have been one of the major concerns for neuroscientists. Thus, strong antioxidant is promising candidate as a drug because evidence has suggested involvement of oxidative stress in the progress of ischemia, amyotrophic lateral sclerosis, Alzheimer's disease, Huntington's disease, and Parkinson's disease. Cabegoline, a dopamine D2 receptor agonist, is an antiparkinson drug, and has been proposed as a potent antioxidant in mesencephalic neurons against oxidative stress, however, influence on cortical neurons expressing D2 receptor was not shown. Here, we showed positive effect of cabergoline on survival of cultured cortical neurons exposed to H2O2 in dose-, pretreatment time-dependent manner. Pretreatment with cabergoline abolished activation of cortical ERK evoked by H2O2 addition, and protective action by blockade of the signaling was confirmed. When focusing on the excitotoxicity(because increased extracellular glutamate, an excitatory transmitter, by oxidative insults contributes to neuronal cell death and stimulates ERK), significant downregulation of extracellular glutamate and upregulation of glutamate transporter(EAAC1 and GLT-1)appeared after cabergoline treatment. We demonstrate that cabergoline protects cortical neurons via decreasing concentration of glutamate due to increased expression of glutamate transporter.

アルギニンメチル化が及ぼす細胞内オルガネラへの影響

Cumulative of reports have shown the importance of ER stress in pathology of neurodegenerative diseases, such as Alzheimer's disease, Parkinson disease, etc. These studies indicate that the cellular events in response to ER stress should relate to the pathology of neurodegenerative diseases. To this aim, we investigated the altered genes in SK-N-SH cells in the condition of tunicamycin-induced ER stress by gene fishing method. As the result, we found that Protein arginine N-methyltransferase 1, PRMT1, is up-regulated in SK-N-SH cells under ER stress. Based on this result, we addressed the following issues;1:Can ER stress increase the protein level of PRMT1? 2:What kind of ER stress pathways is involve in the expression of PRMT1? 3:Can ER stress affect the localization of PRMT1? Our results elucidated that several ER stress pathways induced PRMT1 expression, some of which affected the subcellular localization of PRMT1. Next, to examine the function of PRMT1 in the ER stress response, we downregulated the expression of PRMT1 by RNAi. When we analysed the organelle localization and function in the PRMT1 knockdown cells, the localization of Golgi apparatus was altered and the induction of GRP78 by tunicamycin was severely impaired. These results suggested a novel pathway via protein methylation that mediates organelle stress to the nucleus, possibly involved in the pathogenesis of neurodegenerative diseases.
