ストレス性記憶障害のモデル系:繰り返しLTP後シナプス新生のデキサメタゾンによる阻害

私たちは、培養海馬切片にLTPを繰り返し誘発すると、単発のLTPとは異なりシナプス新生を伴う長期的シナプス強化が起こることを報告している。この構造可塑性現象(RISE;Repetitive-LTP-Induced Synaptic Enhancement)は、多くの点で記憶の固定過程のガラス器内再現と期待される。この系を用いて、ストレスによる記憶障害の細胞過程を解析するため、glucocorticoid(GC)の投与がRISEにどのように干渉するかを検討した。生後6-8日のマウス・ラット新生仔から海馬切片を作成し、2週間以上培養すると、本来の海馬内回路を構築して安定する。この安定培養切片にadenylyl cyclase活性化剤forskolin(20μM、20min)を投与すると、樹状突起棘の拡大を伴うLTPが誘発されるが、24hr以内に消失する。しかし、このLTP誘発を3-24hrの間隔をおいて3回以上繰り返すと、数日かけて発達し3週間以上持続するシナプス強化が起こる。海馬CA1錐体細胞の一部でYFPを発現する遺伝子改変マウスから安定培養切片を作成し、RISEの進行過程を同一細胞同一部位の継時的観察によって追跡すると、次のような段階を経て棘新生が進行する。1)棘は、常時発出と退縮を繰り返す動的平衡状態にある(ゆらぎの平衡)、2)RISE生起刺激後、発出率・退縮率がともに上昇する(ゆらぎの増大)、3)退縮率が先に常時レベルに復帰し、棘数が増す(ゆらぎのバイアス)、4)発出率も常時レベルに復帰し、新たな動的平衡状態に入る。GC受容体特異的agonistとして、dexamethasone(1-100nM)をRISE生起刺激12hr後から24hr投与したところ、刺激10日後の段階でRISE成立は阻害されていた。そこで上記の棘動態に着目して経時的観察を行うと、「ゆらぎの平衡」には影響がないが、「ゆらぎの増大」が阻害されていることがわかった。ゆらぎの変化は、細胞骨格制御因子の調節によることが先行研究で示唆されていることから、GCはこの調節を変調させていると考えられる。

新生仔期フェンサイクリジン連続投与による樹状突起スパイン消失に対するドネペジルによるシナプス再構築

We have previously shown that administration of donepezil after kainic acid-induced morphological changes in dendrites modifies dendritic spine density in the hippocampal and amygdaloid neurons. In the present study, we assessed the neurotoxic effects of neonatal subchronic administration of phencyclidine(PCP), a non-competitive antagonist of the N-methyl-D-aspartate(NMDA)receptor, on the dendritic structure of hippocampal and amygdaloid neurons and a possible role of donepezil, as a sigma-1 receptor agonist, in spine remodelling by using Golgi-Cox staining. Neonatal(age, 7-days)Long-Evans rats were administered PCP(10 mg/kg, s.c.)for 5 days. One week after the last administration, we assessed the effect of repeated(2-week)donepezil(3 mg/kg, i.p.)administration on the dendritic structure, as well as behavioral changes in adulthood. A morphological analysis revealed that subchronic PCP administration decreased dendritic spine density in amygdaloid pyramidal neurons. Donepezil treatment induced the recovery of the decreased dendritic spine density in the amygdala, caused by subchronic PCP administration. These findings suggest that donepezil may promote spinale reorganization in amygdaloid spiny neurons through sigma-1 receptor activity.

Mutations in the Tsc1 or Tsc2 genes cause tuberous sclerosis complex(TSC). Tsc1 and Tsc2 proteins form a complex that inhibits mammalian target of rapamycin complex 1(mTORC1)signalling through Rheb-GTPase. We found that Tsc2+/- neurons showed impaired spine synapse formation, which was resistant to an mTORC1 inhibitor. Knockdown of mTOR also failed to restore these abnormalities, suggesting mTORC may not participate in impaired spinogenesis in Tsc2+/- neurons. To address whether Rheb activation impairs spine synapse formation, we expressed active and inactive forms of Rheb in WT and Tsc2+/- neurons, respectively. Expression of active Rheb abolished dendritic spine formation in WT neurons, whereas inactive Rheb restored spine synapse formation in Tsc2+/- neurons. Moreover, inactivation of Rheb with farnesyl transferase inhibitors recovered spine synapse morphogenesis in Tsc2+/- neurons. In conclusion, dendritic spine abnormalities in TSC neurons may be caused through activation of Rheb, but not through of mTORC1.
