Multivesicular body(多胞体、MVB)は一重膜の胞状構造をしており、内部にintraluminal vesicle(ILV)と呼ばれる直径約40-100 nmの小胞を多数含んだ細胞内小器官である。主にエンドソームを起源として形成されるが、ゴルジ体経由で形成される経路も知られている。近年、MVBがアルツハイマー病をはじめとした神経変性疾患の病理組織で多数観察されることが報告されているものの、中枢神経系におけるMVB形成の生理的意義は完全には理解されていない。我々は、小胞体機能障害によって小胞体内腔に異常タンパク質が蓄積する、所謂、小胞体ストレス状態の際にMVBが増加することを見出した。神経芽細胞腫SK-N-SHやグリオブラストーマU251MGに小胞体ストレス誘導剤ツニカマイシンやサプシガルジンを処理し、3時間後に後期エンドソーム/MVBマーカーであるCD63で免疫染色を行った。コントロールの細胞と比較して、小胞体ストレスを負荷した細胞ではCD63陽性小胞の数および大きさが増した。透過型電子顕微鏡で細胞を観察したところ、ILVを複数含む大型のエンドソーム、すなわちMVBと考えられる構造物が小胞体ストレス時にコントロールと比べ2倍ほど増えていた。一部のMVBは、その内腔に含まれるILVをエクソソームとして細胞外に放出することが知られている。小胞体ストレス後のエクソソーム放出量を調べたところ、コントロールより約2倍増加していた。そこで、小胞体ストレスによって分泌亢進するエクソソームに含まれるタンパク質を質量分析によって解析した。現在、同定したタンパク質がエクソソームに搭載されて分泌される意義について検討中である。 | | 2C-一般1-2 In vivo regulation of GSK3β activity unveiled by the quantitative measurement of its phospho-isotypes 久永 眞市1,Krishnankutty Ambika1,木村 妙子1,斎藤 太郎1,青柳 共太2,浅田 明子1,安藤 香奈絵1,今泉 美佳2,石黒 幸一3 1首都大学東京院・理工・生命科学,2杏林大学医学研究科生化学,3順天堂大学医学部
Glycogen synthase kinase 3β(GSK3β)is a multifunctional protein kinase, which is involved in cell proliferation, survival, development and differentiation. GSK3β is also associated with diseases such as diabetes and Alzheimer's disease. Therefore, it is important to understand the regulation mechanism of its in vivo activity. GSK3β is thought to be constitutive active by autophosphorylation of Tyr216, and inactivated by phosphorylation at Ser9 downstream of many signaling pathways. The kinase activity of GSK3β has been evaluated by inhibitory phosphorylation at Ser9, but it does not measure the the kinase activity properly. Here, we applied the Phos-tag SDS-PAGE technique, which separates proteins depending on phosphorylation states, to the analysis of phospho-isotypes of GSK3β in cells and brains. There were three phospho-isotypes in GSK3β;double phosphorylation at Ser9 and Tyr216, single phosphorylation at Tyr216 and the nonphosphorylation, Active GSK3β with phosphorylation at Tyr216 was most abundant, half or more of total GSK3β. While increase in phospho-Ser9 was detected in insulin-treated cells by immunoblotting with anti-phospho-Ser9 antibody, the increase was a minor fraction of total GSK3β and most GSK3β remained as an active phospho-isotype. Adult mouse brains showed highly active GSK3β with a little Ser9 phosphorylation, and the phospho-isotypes of GSK3β changed depending on the regions of brain, age, sex and disease condition. These results indicate that the Phos-tag SDS-PAGE method provides the simple and appropriate measurement of in vivo active GSK3β and the activity is regulated independently on phospho-Ser9. |
