脳梗塞病態に対する急性期運動療法の脳浮腫抑制効果:ナトリウム/プロトン交換輸送体発現抑制との関連

【緒言】リハビリテーションとして実施される運動療法は、脳卒中後遺症の改善効果を有するとして広く実行されている。しかしながら、運動療法の効果発現メカニズムの分子基盤はよく分かっていない。本研究では、右中大脳動脈一過性(90分)閉塞tMCAOによる脳梗塞モデルラットに対してトレッドミルによる歩行運動を負荷し、その効果と分子基盤について検討した。【方法】tMCAO翌日に小動物用MRIにより脳梗塞容積を計測し、一定範囲内のものをトレッドミルにより、第2、3日目に4m/sec、第4日目に6m/sec、各10分間の歩行運動をさせた。5日目に、再度MRI撮像した後、灌流固定し組織染色に供した。脳浮腫の程度の指標とするため、湿重量と乾燥重量を測定した。脳組織からtotal RNAを調製し定量的RT-PCR(qPCR)を行った。一部のラットは30日間飼育し、運動機能を評価した。運動負荷後に血液を採取し、血中コルチコステロン濃度を測定した。コルチコステロンの経口投与、抗ミネラルコルチコイド薬スピロノラクトン、抗グルココルチコイド薬ミフェプリストンの皮下投与を行った。培養グリア細胞に対するコルチコステロンの効果を調べた。【結果】運動負荷は約10%脳浮腫容積を減少させ、血中コルチコステロンの濃度を軽度上昇させた。コルチコステロンの経口投与は運動負荷と同程度に浮腫を軽減した。運動負荷ラットに、スピロノラクトンまたはミフェプリストンを皮下投与すると、運動による浮腫軽減効果は失われた。運動負荷はアクアポーリン4と各種NHEの発現を抑制した。免疫組織染色により、マイクログリアおよびNG2グリアのNHE1高発現を認めた。コルチコステロンは、培養グリア細胞においてもNHE1の発現を抑え、細胞を縮小させた。【考察・結論】脳梗塞後の脳浮腫発生には、NHE1の発現上昇が関与しており、コルチコステロンはNHE1発現を抑え、細胞浮腫を軽減することで、脳浮腫を改善すると考えられた。運動負荷の脳浮腫軽減効果は少なくとも一部は、コルチコステロンの分泌量を増加させることによってもたらされると考えられた。

Sulfated N-glycans modulate myelination in the peripheral nervous system

Highly specialized glial cells wrap axons with a multilayered myelin membrane in vertebrates. Myelin serves essential roles for a healthy nervous system to function. Axonal degeneration is the major cause of permanent neurological disability in primary diseases of myelin. Sulfated glycans are highly antigenic and the epitopes for auto-antibodies in peripheral demyelinating neuropathy. However, the role of glycans in myelin remains largely unknown. Here, we report that sulfated N-glycans are involved in peripheral nervous system(PNS)myelination. PNS myelin contained highly abundant sulfated N-glycans on glycoproteins. Major sulfated N-glycans in porcine and mouse PNS myelin were identified, and revealed that the 6-O-sulfation of N-acetylglucosamine(GlcNAc-6-O-sulfation)was highly conserved in PNS myelin between these species. P0 protein, the most abundant glycoprotein involved in PNS myelin compaction, had GlcNAc-6-O-sulfated N-glycans abundantly. Mice deficient in N-acetylglucosamine-6-O-sulfotransferase-1(GlcNAc6ST-1)failed to synthesize sulfated N-glycans and exhibited abnormal myelination and axonal degeneration in the PNS. Thus, GlcNAc6ST-1 modulates PNS myelination and myelinated axonal survival through the GlcNAc-6-O-sulfation of N-glycans on glycoproteins. These findings may provide novel insights into the pathogenesis of peripheral neuropathy.

中枢神経系のグルタチオン維持機構に対するさしま茶抽出物の効果の研究

[目的]脳の酸化的ストレスに対する防御にはグルタチオン(GSH)が重要であり、アストロサイトが中枢神経系の主要な抗酸化機構を担っていることが示唆されている。神経細胞のGSHの維持にはアストロサイトからのシステインの供給が不可欠である。茶葉に含まれるカテキン類にアストロサイトの抗酸化活性を高める効果があることが知られている。そこで、さしま茶葉抽出物をアストロサイトに添加して、GSH維持機構に対する効果を調べた。さらに、加工品であるさしま茶飲料との比較を行い、成分分析を行った。[方法]さしま茶葉より熱水抽出した成分、さしま茶飲料を真空凍結乾燥し、生理的食塩水に溶かした。マウス新生児大脳よりアストロサイトを調製し、10日間培養し、実験に使用した。[結果]さしま茶葉抽出物を添加すると、GSHは添加後3時間より上昇し、時間の経過とともに著しく増加し、さしま茶抽出物の濃度依存的にGSH量の増加がみられた。GSHの前駆アミノ酸であるシスチンの取り込み活性もさしま茶抽出物の濃度依存的に増加することがわかった。さしま茶葉の抽出物は著しいGSHの増加をもたらしたが、さしま茶飲料の効果は弱いことがわかった。さらに、さしま茶の成分分析を行ったところ、さしま茶葉とさしま茶飲料で成分に差があることがわかった。クロロゲン酸がさしま茶葉の熱水抽出液に含まれていたが、飲料品になると減少した。また、抗酸化作用が報告されているフラボノイド系ポリフェノールがさしま茶にも含まれていることがわかった。[考察]GSHの前駆アミノ酸であるシステインは細胞外のシスチンに大きく依存していることが知られているので、さしま茶抽出物によるGSHの増加は、亢進したシスチン取り込みによって前駆アミノ酸であるシステインの供給が増えたためであると考えられる。さしま茶抽出物によるアストロサイトのシスチン取り込み活性、GSH量の増加は神経細胞の保護を増強している可能性がある。さしま茶成分は緑茶カテキンに関する既報研究とほぼ合致する結果が得られたが、カテキンの構成比については必ずしも当てはまらなかった。さしま茶でも試料形態によってそれらの構成比が異なり、飲料としての機能性に影響を及ぼす可能性が示唆された。

Effects of the anti-Large myelin protein zero antibody on the rat lysolecithin-induced peripheral nerve demyelination

Large myelin protein zero(L-MPZ)is a recently identified novel isoform of P0, in which extra 63 amino acids are added at the C terminus by stop codon readthrough mechanism. Antibodies against L-MPZ were often found in the sera from the patients with chronic demyelinating polyneuropathy(CIDP), although pathophysiological role of these antibodies are still uncertain. Intravenous IgG Kenketu glovenin-I(IVIg)prepared as a mixture of IgG from thousands of healthy donors contains a low dose of anti-L-MPZ antibodies. To determine the relevance of anti-L-MPZ antibodies to neuropathic conditions, we examined rodent demyelinating nerves in the presence and absence of IVIg.
ICR mice were intraneurally injected with 1% Lysorecithin into the left sciatic nerves, and they were intravenously injected with either IVIg or saline(as a control)24 hours later. On days 7 and 14 after lysolecithin injection, sciatic nerves were resected, and examined by immunofluorescent study. Evaluations of demyelinating areas and classification of infiltrated macrophages were performed.
By western blotting using IVIg, only 36 kDa band of L-MPZ was detected in rat sciatic nerve homogenates. Injected human IVIg bound to myelin debris in the demyelinated area, which was similar to the patterns by anti-L-MPZ antibodies. Infiltrated macrophages were observed in the demyelinating region and surrounding areas. Macrophages were classified by their phenotypes, by labeling with anti-CD68 and anti-Arginase1. On day 7 after lysolecithin injection, demyelinating areas of IVIg injected group were smaller than those of saline injected group.Numbers of CD68+Arginase1+ M2 macrophages were increased in IVIg injected group, while CD68+Arginase1- macrophage were increased in saline treated group.
Thus, present results indicate that IVIg treatment has protective effect on chemical demyelinating condition through induction of M2 macrophages. Since human IgG from IVIg directly binds to the myelin debris, may modulate peripheral demyelinating condition.

The majority of excitatory synapses in the mammalian cerebral cortex occur at spines on the dendrites. In primates, the number of dendritic spines rapidly increases after birth, reaches a peak at around the early infancy or mid-childhood period, and then decreases towards the adult level. Abnormalities of the processes of spinogenesis and pruning are implicated in several psychiatric disorders, such as autism spectrum disorder(ASD), and schizophrenia. In ASD, excess spine formation or incomplete pruning may occur, which leads to increased spine numbers.We investigated the normal processes of spine formation/pruning in the cerebral cortex of the common marmoset as a primate model. Our previous studies showed similarities and differences of developmental profiles of basal dendrites and spines of layer-III pyramidal cells among the six cortical areas(V1, TE, area 12, area 9, area 14r, area 24), and reported that all cortical areas showed overshoot-type spine formation with peaked at 2-3 month old. We have developed ASD model of the marmoset by expose to valproic acid(VPA)during gestation period. The VPA marmoset has a deficit in communication and perseveration, which are core features of ASD(Yasue et al., 2015). In this study, we measured the thickness of cortices, the density of cortical neurons, and the analyzed the processes of spine formation/pruning in the six cortical areas of the model animals. On 2, 3, and 6 month old, the thickness of gray matter was found to be increased in the VPA-exposed animals compared with that in unexposed(UE)animals. The density of cortical neurons was lower in the model animals than that in the UE animals. Layer-III pyramidal cells in the areas were intracellularly injected with Lucifer Yellow in lightly fixed slices, and reacted for Diaminobenzidine. Basal dendrites of more than 150 cells were reconstructed, and their morphological features were analyzed. In the prefrontal cortical areas, we found that insufficient spine pruning of the ASD model compared with UE animal. Search for the molecular mechanisms that underlie the developmental changes will provide further clues for an understanding of pathogenesis of ASD.

Microglial abnormality in the cerebral cortex of a primate model of autism spectrum disorders

Microglia are supposed to be surveyors and scavengers in the brain. Recently, microglial abnormalities have been observed in the brains of patients with neuropsychiatric disorders including autism spectrum disorders(ASD), suggesting that microglia are implicated in the pathophysiology of these disorders. In the cerebral cortex of primates including human, the number of dendritic spines rapidly increases after birth, reaches a peak at around the infancy or mid-childhood, and decreases towards adulthood. In neuropsychiatric disorders, the abnormalities in the number of dendritic spines are one of the pathological findings. In ASD, excess spine formation and imperfect synaptic elimination(pruning)result in increased spine number. To reveal microglial involvements in the pathophysiology of ASD, we have developed ASD model of the common marmoset(Callithrix jacchus)by fetal exposure to valproic acid(VPA). The VPA marmosets have a deficit in social interaction, which is one of features found in ASD patients(Yasue et al., 2015). In this study, we immunostained multiple cortical areas using anti-Iba1 antibody and observed the morphology of Iba1-immunopositive microglia. We performed immunohistochemistry at 2M, 3M, 6M and adult, and investigated the developmental changes of microglia in VPA-exposed marmosets and unexposed(UE)marmosets. In several cortical areas, we found the morphological abnormality of microglia(e.g. decreased density, change in process form)in VPA marmosets compared with UE marmosets. Our results support the microglial abnormalities observed in the brains of ASD patients. The VPA marmosets are useful primate model of ASD and further analysis using this model would lead to understand the microglial roles in the pathophysiology of ASD.

Molecular pathogenesis of dysmyelination in lysosomal storage disease, Niemann-Pick disease type C.

A large amount of cholesterol is required for myelination in the developing nervous system. Lysosomal storage diseases(LSDs)are childhood-onset inherited neurodegenerative diseases caused by defects in lysosomal function required for metabolism of lipids, proteins or glycosaminoglycans. Some of LSDs show pathological features of myelin defects;however, their underlying mechanisms are poorly understood. Here, we examined molecular pathology of NPC1-deficient mouse, an animal model of Nieman-Pick disease type C(NPC), one of the LSDs associated with abnormal lysosomal cholesterol trafficking. During early postnatal development, NPC1-deficient mice showed impaired myelination indicating dysmyelination. Activities of some signaling molecules for oligodendrocyte differentiation were significantly reduced in NPC1-deficient mouse brain. Moreover, in primary cultures of oligodendrocyte precursor cells, inhibition of lysosomal cholesterol trafficking affected oligodendrocyte differentiation. Our results suggest that lysosomal cholesterol trafficking plays a critical role for oligodendrocyte differentiation and myelination in the CNS.
